A schematic model for ZJW extracts in inhibiting 5-HTR1D and attenuating Wnt/β-catenin signaling pathway. Wnt/β-catenin signaling is activated by the binding of Wnt ligand to Frizzled receptor and LRP5/6 co-receptors, leading to the recruitment of Dvl and destruction complex to the membrane, which inactivates destruction complex, causing accumulated β-catenin to enter nucleus and activate target gene transcription (red arrows). 5-HTR1D may play a role in promoting the Wnt/β-catenin signaling at upper stream, leading to stabilization of β-catenin (red arrows). The effect of ZJW extracts on Wnt/β-catenin resembles that of GR127935 (GR), an antagonist of 5-HTR1D, in inhibiting (green arrows) the Wnt/β-catenin signaling through enhanced β-catenin degradation, thus prohibiting the signaling transduction
