Inhibition of TNF-α-induced MMP-9 activation by SSeo in HASMCs. (A) HASMCs were pretreated for 1 h with different concentrations of SSeo, followed by incubation with TNF-α for 24 h. The culture medium was collected and analyzed for gelatinolytic activity by zymography. (B)In vitro activity of MMP-2 and −9 in cell culture supernatant was measured using a MMP gelatinase activity assay kit. The biotinylated gelatinase substrates were cleaved by active MMPs in the samples, and the fragments were added to a biotin-binding plate. The digested but unbound fragments were removed by washing. Data are mean ± SD from three independent experiments and are presented as fold change compared with untreated control cells (*, p < 0.05 vs. untreated control; #, p < 0.05 vs. TNF-α-treated HASMCs).
